After the light source emits strong light and stabilizes, the eyes fully adapt to the darkroom and begin to observe the specimen. 3, to prevent damage to the eyes caused by UV, and then adjust the light, you should bring protective eyes. 4, the inspection time to 1-2h is appropriate, if more than 90min, ultra-high pressure mercury lamp light intensity gradually decreased, fluorescence weakened; specimens by ultraviolet radiation 3 ~ 5min, the specimen fluorescence is also significantly reduced; so no more than 2 ~ 3 . 5, Fluorescent light source life is limited, specimens should be checked in order to save time and protect the light source. When it is hot, fans should be turned on or the air conditioners must be cooled to cool down. Change the new bulb should record the time from the beginning. After the lamp is extinguished, it is intended to be used. The lamp can be ignited only after the lamp has cooled sufficiently. Avoid lighting the light source several times during the day. 6. Immediately after specimen staining, the fluorescence will gradually decrease over time. If the specimen is stored in a polyethylene plastic bag and stored at 4 degrees, the fluorescence decay time can be retarded. 7. Judgment criteria for fluorescence intensity; generally divided into four levels, 1: no or weak fluorescence visible, 2: visible only visible fluorescence, 3: visible bright fluorescence, 4: visible bright fluorescence.
