**Purpose of Use:** This kit is designed for the quantitative determination of human S100 calcium-binding protein A8/calcium granulin A (S100A8) in human serum, plasma, and other related biological samples. It provides a reliable and accurate method for measuring S100A8 levels, which is useful in clinical diagnostics and research settings.
**Experimental Principle:** The assay is based on the double-antibody sandwich enzyme-linked immunosorbent assay (ELISA) principle. A microplate is pre-coated with a specific monoclonal antibody against human S100A8. After incubation with the sample, the target antigen (S100A8) binds to the immobilized antibody. A second HRP-conjugated antibody is then added, forming a complex of antibody-antigen-enzyme-labeled antibody. Following thorough washing to remove unbound components, a TMB substrate is introduced. Under the catalytic action of HRP, TMB turns blue, and upon addition of an acidic stop solution, it changes to yellow. The intensity of the color is directly proportional to the concentration of S100A8 in the sample.
The optical density (OD) is measured at 450 nm using a microplate reader. By comparing the OD values of the test samples to a standard curve generated from known concentrations of S100A8, the exact level of S100A8 in the original sample can be accurately determined. This method ensures high specificity, sensitivity, and reproducibility, making it ideal for both routine and research applications.
