Sample type (SAMPLE TYPES):

Specimens must be liquid and free of sediment. Including serum, plasma, urine, pleural and ascites, cerebrospinal fluid, cell culture supernatant, tissue homogenate and the like. 1 ml of whole blood can give 0.5 ml of serum or plasma. Each specimen volume collection volume = 100 ul x detection type. Ask the salesperson for instructions before taking the material.

Before collecting the specimen, it must be clear whether the ingredients to be tested are stable enough. Specimens that were tested on the day after collection should be stored at 4 °C for future use. If there are special reasons, the specimens should be collected periodically, and the specimens should be dispensed in time and stored at -20 °C or -70 °C. Avoid repeated freezing and thawing. Specimens can be stored for 48 hours at 2-8 °C and for 1 month at -20 °C. -70 degrees can be stored for 6 months. Some hormone samples require the addition of aprotinin.

3. Serum:

EDTA, sodium citrate or heparin should be selected as anticoagulant according to the requirements of the kit. Add 10% (v) anticoagulant (0.1M sodium citrate or 1% heparin or 2.0% EDTA.Na2) for 10-20 minutes. After that, centrifuge for about 20 minutes (2000-3000 rpm). Collect the supernatant carefully. If a precipitate forms, it should be centrifuged again.
4. Urine, chest and ascites, cerebrospinal fluid:
Collect with a sterile tube. Centrifuge for about 20 minutes (2000-3000 rpm). Collect the supernatant carefully. If a precipitate forms, it should be centrifuged again.
5. Cell culture supernatant:
When detecting secreted components, collect them in a sterile tube. Centrifuge for about 20 minutes (2000-3000 rpm). Collect the supernatant carefully. When the components in the cells were detected, the cell suspension was diluted with PBS (pH 7.2-7.4), and the cell concentration reached about 1 million/ml. By repeated freezing and thawing, the cells are destroyed and the intracellular components are released. Centrifuge for about 20 minutes (2000-3000 rpm). Collect the supernatant carefully. If a precipitate forms during storage, it should be centrifuged again.
6. Organize specimens:
After cutting the specimen, weigh the weight. A certain amount of PBS was added, and 1 μg/L protease inhibitor or 50 U/ml Aprotinin (aprotinin) was added to the buffer. The specimen was homogenized by hand or homogenizer. Centrifuge for about 20 minutes (2000-3000 rpm). The supernatant is carefully collected and stored at -20 or -70 degrees. If necessary, the sample can be concentrated and dried. One part of the package is to be tested, and the rest is frozen for use.
